本試驗目的為構築雞隻 W 染色體上可能的性別決定候選基因 chHINTW 慢病毒基 因載體 pLAS2.1w.PeGFP-I2-Puro-HINTW，並感染雞 DT40 細胞以探討HINTW 基因 過表現對 DT40 細胞之影響。試驗利用 chHINTW ORF（open reading frame）序列， 構築 pLAS2.1w.PeGFP-I2-Puro-HINTW 慢病毒基因載體，轉染至 293 細胞製作慢病 毒顆粒後與計算相對病毒力價後感染雞 DT40 細胞，分析 HINTW 基因過表現對 DMRT1 和 MHM 性別相關基因之影響。結果顯示，病毒用量 0、0.3、0.4、0.6、0.8 及 1 μL，並經 puromycin 篩選 48 小時後與 DT40 細胞標準生長曲線相較下之細 胞存活率分別為 0、20、33、45、66、80%（R2 = 0.9893），取最佳細胞存活率的病毒 用量 1 μL之細胞存活率計算相對病毒力價為 2.25x106 R.I.U./ mL。而 DT40 細胞經 帶有 HINTW 基因之慢病毒感染後，細胞內 HINTW 表現較無外源性 HINTW 基因 者顯著提高，DMRT1 基因表現亦顯著上升，但 MHM 基因表現則無影響。後續可利 用 HINTW 基因慢病毒進行in ovo 試驗，以研究 HINTW 於雞隻性別決定機制中所 扮演的角色。
英文摘要
This study was conducted to establish the construct of potential candidate gene of chicken sex determination, HINTW, on lentiviral vector and infect DT40 cell line by HINTW-lentivirus for evaluating the effect of HINTW gene overexpression in DT40 cell line. The results showed that 0, 0.3, 0.4, 0.6, 0.8 and 1 uL lentiviral particle infection with 48 hours selection of puromycin, the DT40 cell survival rates were 0, 20, 33, 45 and 66, 80% (R2 = 0.9893), respectively. The relative virus titer, calculating by best viability group which was 1 uL virus treatment, was 2.25 x 106 R.I.U./ mL. In HINTW-overexpressed DT40 cell group, the expression of HINTW and DMRT1 were significantly higher than control DT40 cell group, but the expression of MHM gene did not show any difference between the two groups. This HINTW-lentivirus system could be used for the further in ovo study to investigate the role of HINTW gene in chicken sex determination.
