臺大李宋豬為小閉鎖族群,在進行近親配種繁衍,出現包括無眼、無虹膜及小眼症等眼睛缺陷之體 表型。本試驗目的在於將無眼李宋豬進行無特定病原化,以利將來在生物醫學應用。試驗前將臺大 豬群進行兩季疾病監控,確認達符合無特定病源之法規規定狀態,再分別在臺大及農科院進行供受 胚豬發情同期化、誘發排卵、沖胚/胚移置及剖產代養等引種工作。胚移置係在臺大動物醫院以外 科手術取2- ~ 4-細胞期眼睛缺陷李宋豬胚,攜回農科院,於 4 h 內移入受胚母豬輸卵管內;剖產/ 代養係將懷孕之李宋母豬於預產期運至農科院,進行帝王切開術剖產,取出仔豬直接引入分娩舍, 由同期化自然分娩之母豬代理哺乳。在胚移置方面,進行兩批次試驗共4 頭供胚豬,獲得11 個4- 細胞、1 個 2-細胞及 21 個 1-細胞期豬胚,在搭配至少 4 個一般豬胚進行移置後,合計3 頭受 胚豬均未懷孕;在剖產/代養方面,共剖產 2 頭懷孕母豬,獲得 4 頭(2 公2 母)雙無眼、1 頭 (母)無虹膜/小眼及 4 頭(1 公3 母)雙無虹膜仔豬。結果顯示眼睛缺陷李宋豬因誘發排卵數 限制,進行 SPF 化引種,以剖產/代養之效率較胚移置者為佳。
The Lee-Song pigs in National Taiwan University (NTU) are a small and closed inbred herd and some appear ophthalmic defects including anophthalmia, microphthalmia and aniridia. The purpose of this study was designed to introduce anophthalmia Lee-Song pigs into specific pathogen free (SPF) farm for biomedical research. Before introduction of embryos and animals, the SPF pathogens in NTU farm were detected twice in two different seasons to reveal their biosecurity conditions comply with SPF guidance. All the donors in NTU and recipients as well as foster dams in Agriculture Technology Research Institute (ATRI) were estrus synchronized to induce ovulation concurrently. The flushed embryos at NTU were transferred (ET) into the fallopian tubes of recipients at ATRI within 4 h. Overall, 11 4-cell, 1 2-cell and 21 1-cell embryos were recovered from 4 donors and ET into 3 recipients, together with at least 4 wild-type embryos in each attempt, but failed to impregnate any recipients. In C-section approach, two pregnant Lee-Song sows were operated and obtained 4 anophthalmia (2 male and 2 female), 1 microphthalmia and aniridia (female) and 4 aniridia (1 male and 3 female). Due to the limited numbers of induced ovulation, we suggest that to obtain Lee-Song piglets with inherited anophthalmia by C-section and nursed by foster dams will be more successful than ET.
