黑酵母(Aureobasidium pullulans)為一常見的真菌,因菌落為褐色或黑褐色,稱之黑 酵母,目前為衛福部之可供食品使用原料之一,其安全性已受肯定。葡聚糖為黑酵母 發酵後之產物,具有免疫調節及抗氧化等多種功效。本試驗之目的,即探討黑酵母液 態發酵後分離之葡聚糖作為主要的研究標的物,分析其生理活性與抗發炎作用。結果 顯示,黑酵母中的葡聚糖可降低 LPS 刺激之小鼠 RAW 264.7 巨噬細胞發炎反應,其 中 0.545 mg/mL 的黑酵母葡聚糖對細胞一氧化氮(NO)產生抑制率達 40%,透過酵 素免疫分析證實,黑酵母葡聚糖可顯著降低其 TNF-α、IL-1β 與 PGE2 的產生。此 外黑酵母葡聚糖同時具有毒殺癌細胞之功能。綜合以上所述,黑酵母發酵產品對於毒 殺癌細胞、抗發炎皆有效果。

英文摘要

Aureobasidium pullulans is a common fungus with brown or dark brown colonies, which also called the black yeast. The black yeast is currently available in that food category for the Department of Ministry of Health and Welfare of Taiwan. β-Glucans produced by A. pullulans, could have a profound impact for immune regulation and anti-oxidation. The purpose of this study is to investigate the biological activities and anti-inflammatory effects of β-glucans produced by black yeast. The results showed that β-glucans could decrease the lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophages. The inhibition rate of nitric oxide (NO) production was 40% in the treatment of 0.545 mg/mL β-glucans. β-Glucans can significantly reduce the level of TNF-α, IL-1β and PGE2 determined by ELISA. In addition, black yeast β-glucan revealed the anti-cancer activity in the vitro test. Our results suggest that the β-Glucan derived from A. pullulans is a potential candidate used for anti-inflammatory and anti-cancer therapy.
