豬小腸粘膜下層(SIS)目前已有商品廣泛使用於臨床中組織重塑的各種適應症, 包括血管和心臟修復。小腸粘膜下層的細胞外基質(ECM)經由去細胞過程處理, 但仍保留基質的天然纖維和多孔性質以及若干基質相關細胞因子。去細胞化旨在 從其 ECM 中除去細胞,從而去除可引起炎症反應或植入時引發免疫排斥反應的 潛在抗原。在這個研究中,我們開發了生產無細胞 SIS-ECM 的方法,並經由分 析DNA 含量,糖胺聚醣含量,代謝活性和組織化學染色鑑定期品質。本製程可生 產具有一致性品質的產品,並且有很好的生物相容性。
Porcine small intestine submucosa (SIS) is commercially available and widely used for tissue remodeling in the clinic for various indications, such as vascular and cardiac repair. The experimental material was derived from SIS through a process where the extracellular matrix (ECM) was decellularized while still retaining the naturally fibrous and porous nature of the matrix, as well as several matrix-associated cytokines. Decellularization was aimed to remove cells from their ECM and thereby removing potential antigens that can cause inflammatory responses or immune-mediated implant rejections. In this work, we developed a process for producing acellular SIS-ECM and analyzed the quality of the productions by DNA content, glycosaminoglycan content, metabolic activity, and histochemical stain. The products manufactured by this process showed a consistent quality with high biocompatibility.
