本試驗探討添加三種抗氧化劑-維生素E(alpha-tocopherol,Vit E)、丁羥基甲苯(butylated hydroxytoluene, BHT)與麩胱甘肽(glutathione, GSH)於冷凍稀釋液對精液解凍後品質的影響。 採取 5 頭公豬之新鮮精液,以 Lactose-egg yolk(LEY)之精液稀釋液稀釋,稀釋最終濃度為 5×108 cell/mL。試驗比較添加 5 mM GSH、1 mM BHT 及 200 μg/mL VitE 濃度加入冷凍稀釋 液作為比較,評估解凍後的精子活力、精子快速前進式活力及精子活力各項移動參數等項目之 差異。試驗結果顯示添加 5 mM GSH 於冷凍稀釋液,精子活力及精子快速前進式活力之均較 200 μg/mL VitE 組差。但 200 μg/mL VitE 組與對照組比對,精子活力及精子快速前進式活力 雖有提升,但統計無顯著差異(P > 0.05),在精子活力各項移動參數方面,精液解凍後體外 培養 5 min 及 6 h,冷凍稀釋液添加 5 mM GSH,精子活力各項移動參數包括 VAP、VSL、 VCL 及 ALH 均呈顯著低現象,但所有指數於上述各組間均無顯著差異(P > 0.05)。說明冷 凍稀釋液添加 5 mM GSH、1 mM BHT 及 200 μg/mL VitE,對解凍後精子的各項活力參數沒 有顯著影響。

This experiment was conducted to investigate the effects of three antioxidants -vitamin E (Vit E), butlated hydroxytoluene (BHT) and glutathione (GSH) were added to freezing extender on quality of frozen-thawed semen in boars. Semen collected from five boars were diluted with Lactose-egg yolk (LEY) extender, which it brought to 5 x 108cell/mL in the final concentration. The effects of addition of 5mM GSH, 1 mM BHT and 200 μg/ml Vit E into freezing extenders were evaluated. The percentage of sperm motility, rapid progressive motility and motility kinetic variables parameters were recorded. The results showed that the percentage of total motility and rapid progressive motility in 5mM GSH supplemented freezing extender groups were lower than in 200μg/ml Vit E. The percentage of total motility and rapid progressive motility in the 200μg/ml Vit E group after thawing were better than the control. However, there is no significant difference between the 200μg/ml Vit E supplemented and control group (P > 0.05). The results showed that the percentage of sperm motion parameters (VAP, VSL, VCL and ALH) of semen cryopreserved in freezing extender after thawing for 5min, 6 hrs were lower in 5 mM GSH group than in the others. However, no differences were found between the percentages of sperm motion parameters (P > 0.05). In conclusion, the addition of 5mM GSH, 1mM BHT and 200μg/ml Vit E to the freezing extender did not result in any improvement in sperm motility parameters.
