中文摘要

研究對黃芩根萃取物進行成份分析與美白保養品功效之評估,並比較萃取製程放大後之影響。將一 批小量(S)與放大 200 倍製程(L)之萃取產物以 DPPH、ABTS 試驗評估萃取物的抗氧化能力, 結果顯示兩者萃取物皆具有劑量正相關清除氧化自由基能力,DPPH 試驗中 S、L 萃取物在 50 μg/mL 濃度分別有 77% 與 74.5% 之清除率,ABTS 試驗中則有 63.4 與 60.8% 之清除率。另 外,將萃取物處理彈力蛋白酶(elastase)以分析抗老化功效,S、L 萃取物在 200 μg/mL 濃度也 均可抑制 47%、52.7% 酵素活性,其功效相當。在抑菌能力方面,S 萃取物處理Staphylococcus epidermidis,其抑菌生長的最低濃度(MIC)1.875 mg/mL,高於 L 萃取物之 0.625 mg/mL 約 3 倍。 進一步以高效能液相層析儀(HPLC)分析萃取物裡之黃芩根活性物質,得知 S 萃取物之 baicalin 含量比例占 45.11% 為 L 萃取物(16.43 %)2.7 倍,但 baicalein 含量(1.06%)卻低於 L 萃取 物(3.16%)3 倍,由此推測萃取製程放大後,產出之萃取物成份比例改變,可能影響抑菌效果。 以上實驗結果得知萃取物具有良好抗氧化、老化及抗菌功效,在運用於美妝保養品開發時,必須考 慮量產製程上的一些影響因子並加以最適化調整。

英文摘要

The active components and functional effects of Scutellaria baicalensis (S. baicalensis) root extracts from large scale extraction were evaluated in this study. Both batches of extracts produced in small (S) and 200 times large manufacture (L) were analysis anti-oxidant property by using DPPH and ABTS assay. The result showed scavenge rate of 50 μg/mL S、L extracts were 77、74.5 % for DPPH assay, and 63.4、60.8 % for ABTS assay, both of extracts were able to scavenge free radicals in dose-dependent manner. In addition, the anti-aging property of extracts was evaluated with the effect on the elastase activity. S and L extracts in 200 μg/mL inhibited elastase activity for 47 and 52.7 %, respectively, demonstrating the similar efficacy between both extracts. In terms of antibacterial ability, the minimum inhibitory concentration 1.875 mg/mL of S extract for Staphylococcus epidermidis was 3 times as high as L extract 0.625 mg/mL. Furthermore, the active components of S. baicalensis root extracts were determined by high performance liquid chromatography (HPLC), showing baicalin content proportion of S extract (45.11 %) was 2.7 times higher than L extract (16.43 %), but baicalein content proportion (1.06 %) 3 times lower than L extract (3.16 %). This result in different anti-bacterial capability was inferred to the ratio change of component content in extracts when manufacture was enlarged. Taken together, S. baicalensis root extracts have effects on anti-oxidation, anti-aging and anti-bactericidal. It may be still required to consider the production process for optimization, as applying to develop skin-care cosmetic products.
