食品在加熱、烘乾及煙燻過程中會產生多環芳香族碳氫化合物(polycyclic aromatic
hydrocarbons, PAHs),部分PAHs已証實可能具致癌性,其中苯芘(Benzo[a]pyrene)可作
為食品中具可能致癌性PAHs存在及其效應之指標。歐盟執委會208/2005號規範特定食品中苯芘最大限量,其中食用油脂為2.0 μg/kg。本研究以HPLC建立食品油脂中14種PAHs含量之檢驗方法。食用油脂以isohexane/t-butyldimethylether(95 : 5, v/v)溶解後,經HR-P固相萃取匣淨化,最後以高效液相層析儀附螢光檢出器進行PAHs含量之檢測。高效液相層析部份,係以Supelcosil LC-PAH層析管柱進行分離,以乙腈及水為移動相進行梯度沖提,於螢光檢出器激發波長290∼314 nm及放射波長398∼511 nm偵測。14種PAHs之標準曲線判定係數皆大於0.999,於食用油脂中添加5 μg/kg之PAHs,其回收率介於60∼106%,檢出限量為0.04∼3 μg/kg,定量極限為0.2∼10 μg/kg。應用本研究所建立之方法檢測市售食用油脂橄欖油等20件,結果有16件檢體檢出含有PAHs,13件檢出苯芘,含量範圍為0.20∼0.73 μg/kg,均低於歐盟規範油脂之最大限量2.0 μg/kg,顯示無安全顧慮。

英文摘要

Polycyclic aromatic hydrocarbons (PAHs) form during food processing or cooking. Some PAHs were considered as carcinogens. Benzo(a)pyrene was used as a marker for the occurrence and effect of carcinogenic PAH. In Commission Regulation (EC) No 208/2005, maximun level of benzo(a)pyrene in edible oil was 2.0 μg/kg.
A method to analyze the contents of 14 PAHs in edible oil was developed in this study. Benzo(a)pyrene was extracted from edible oils using isohexane/t-butyldimethylether (95:5, v/v) and cleaned up with HR-P cartridge.The extracts were analyzed by high performance liquid chromatography using with fluorescence detector (HPLC/FL). The Supelcosil LC-PAH column was used for the separation in HPLC. The LC was affected with a gradient elution using acetonitrile and water as mobile phase. Excitation and emission wavelengths of the detector were 290 - 314 nm and 398 - 511 nm, respectively. All the linear regression coefficients standard curves of PAHs were higher than 0.999. The recoveries of PAHs (5 μg/mL) spiked in edible oil were between 60 - 106%. The limit of detection ranged 0.04 - 3 μg/kg and the limit of quantification was 0.2 – 10 μg/kg. This method was well qualified for the performance criteria of analysis that is requested by EC. The 13 items of the benzo(a)pyrene
were detected in 20 various edible oils with a concentration range of 0.20 - 0.73 μg/kg.
