採用表面電漿共振檢測方式之生物感測器無須事先標定待測物,便具有高靈敏度、可定性/定量/動態分析生物分子交互作用,以及進行大量平行篩檢等優點,因此,相較於須進行螢光或輻射標定等之生物檢測法,有更好發揮之潛力,而研究者結合多層膜之菲涅耳(Fresnel)方程式及菱鏡斜邊之折射定律,計算出不同形狀之菱鏡耦合表面電漿共振角度反射光譜,以及完整分析不同形狀稜鏡耦合元件對薄膜表面電漿共振反射光譜與共振角檢測靈敏度之影響,並得到等邊菱鏡對應之共振角靈敏度皆比半圓稜鏡大。另外,等邊菱鏡之底角愈小則對應共振角靈敏度愈大,然而就系統檢測解析度而言,不同形狀菱鏡之間並無明顯差異,但對於不同偵測目的選擇適當靈敏度之菱鏡耦合元件仍是必要的。

英文摘要

Biosensors based on surface plasmon resonance detection method without prior labeling of the analytes have many advantages such as high sensitivity and capabilities of qualitative / quantative / parallel / dynamic analysis of biomolecular interactions. In this work, multilayer Fresnel equations and refraction law of the prism are combined to calculate the angular reflection spectrum of the film surface plasmon resonance for the semicircle and equilateral triangle prism coupling elements. Sensitivities of resonance angle are analyzed between different shapes of prisms. It is found that the triangular prism with bottom angles of 40°~80° has larger sensitivity than the semicircular, and the triangular prism with smaller bottom angle has larger sensitivity. The system detection resolutions are the same for different prisms. However, an appropriate prism coupling element with specific sensitivity is still required for different detection purposes.
