本研究利用中草藥阿膠及明膠為創傷敷材之主要基材,搭配三黃與石膏,進行中草藥結合明膠敷材設計製作及物、化性分析、中草藥結合明膠敷材藥物載體實驗以及生物相容性實驗,透過三種不同材料濃度(阿膠暨明膠比例為3:7、5:5、7:3)及三種不同材料濃度(15%、20%、25%),並觀察其應用於創傷敷材之特性及潛力。研究結果發現,加入其他材料不影響阿膠與明膠的化學結構且材料之玻璃轉換溫度均在材料穩定性範圍;濃度20%拉伸距離為最高值,濃度25%擁有較佳的透氣度;透過最佳化參數實驗,遴選材料比例(阿膠:明膠50:50),材料濃度20%及25%進行後續實驗,其中以濃度20%擁有較為親水之性質。藥物包覆率以濃度25%為最高,藥物釋放率以濃度20%為最佳,另外,兩實驗之組膨潤度皆有近原體積6倍之表現;材料濃度20%及25%之創傷敷材皆可使細胞顯著增生,證明其具有良好的生物相容性。綜合上述實驗結果,以材料比例(阿膠暨明膠比例5:5),材料濃度20%,所製備之創傷敷材具備成為臨床傷口敷材之良好特性及潛力。

英文摘要

This study utilized Chinese herbal medicine Colla Corii Asini and gelatin, as the basic material of wound dressing. The wound dressing also used the natural medicine of San-Huang and gypsum, then through the experiment of material design and physical-chemical properties analysis, experiment of as adrug carrier and the material biocompatibility. Study use three different concentration of the material by (gelatin and gelatin ratio of 3:7, 5:5, 7:3) and three different material concentration (15%, 20%, 25%), and observe the characteristics and potential of wound dressing material to application. The results showed that by adding other materials, the chemical structure of Colla Corii Asini and gelatin is not affected. And adding different herbs can cause a transition in the glass material temperatures that is not in the range of material stability. The concentration of 20% elongation of the distance the highest value. For air permeability, the concentration of 25% has better air permeability. Through the above parameter optimization experiments, the materials that were used had a concentration of 20% and 25% for subsequent experiments, in which the concentration of 20% showed a more hydrophilic nature. For coating rate, the concentration of 25% has the highest rate of the cladding. Release rate of 20% concentration is the best. Swelling ratio test, two experimental groups individually had nearly six times the performance of the original volume. The material concentration of 20% and 25% of the wound dressing material can be a significant proliferation of the cells, which proves it has a good biocompatibility. Taking these experimental results, the study using material (gelatin and gelatin ratio of 5:5), material concentration 20%, the materials have good characteristics and potential to become a clinical wound dressing material.
