本研究由防風Saposhnikovia divaricata (Turcz.) Schischk.中抽取純化特有之鎮痛、降血壓之活性成分cimifugin作為指標成分,並建立防風藥材中cimifugin的高效液相層析定量分析模式;其分析條件以CH3CN-H2O(18:82)為移動相,流速為1.0mL/min,以LiChrospher 100RP-18e(4*250mm)為分析管柱,以波長220nm檢測,在40℃下進行分析。防風中cimifugin的最佳抽取條件為以20倍量70%甲醇水溶液經超音波震盪40分鐘可得最高產率。利用上述高效率液相層析法定量巿售防風藥材中cimifugin的含量,結果cimifugin含量在2.66*10-2%至0.19*10-2%之間。
Cimifugin, one of the principal analgesic and anti-hypertensive componentof Fang-Feng, was isolated from the root of Saposhnikovia divaricata. Reversed-phasehigh-performance liquid chromatography was employed to determine the contents ofcimifugin in Fang-Feng. The separation was performed on a LiChrospher 100 RP-18ecolumn (4*250 mm) by isocratic elution with acetonitrile-water (18:82) as the mobilephase at a flow-rate of 1.0 mL/min, the temperature being kept at 40℃ and thedetection set at 220 nm. Methylparaben was used as the internal standard. Theregression equation revealed linear relationship between the peak-area ratio (markersubstance/internal standard) and concentration. We found that Fang-Feng extractedwith twenty-fold of 70% aqueous methanol under sonication for 40 minutes gives thebest yield of cimifugin. The contents of cimifugin in commercially availableSaposhnikoviae Radices have been determined and the amount was found to vary from2.66*10-2% to 0.19*10-2%. This HPLC method can rapidly and easily quantify thecimifugin present in Fang-Feng and can be used for quality control of this commercialcrude drug.g.
