     本研究目的在於藉由瞭解幾丁聚醣、膠原蛋白、明膠摻合膜材的表面性質與微構形對C3A人類肝癌細胞行為表現的影響,希望能找出加速細胞生長速率或引導細胞分化的條件,以應用於組織工程的範疇中。研究中先利用光蝕刻技術製作具微構形之不?袗?材母模,再配合軟蝕刻技術將微構形轉印於高分子膜材上。將C3A細胞植入於間距為200μm及深度為80μm的表面微構形膜材上,細胞在數分鐘內即完成貼附動作,不再懸浮,且落點大都聚集於構形的凹槽內,在凸處只有幾顆零星的細胞分佈。隨著時間的增加,在凹槽邊緣的細胞會進行移動與形態上的分化,並往上爬升,佔據構形凸處未存有細胞的區域。觀察細胞在不同膜材上的生長表現,發現生長在具表面微構形膜材上的細胞比在T-flask上的細胞能具有更快的生長速率。在此證明膜材上的微構形會影響細胞整體的分佈,進而影響細胞間的溝通方式,而導致細胞呈現不同的行為表現。

英文摘要

     Guiding the cell growth on biomaterials is of importance in the practice of tissue engineering. The effect of the surface micropatterns on the cell culture of C3A cells on the chitosan-collagen-gelatin (CCG) blended membranes was studied. Stainless steel masters with micropatterns were prepared by photolithography. The micropatterns were then transferred to the CCG membranes through the technique of soft lighography. When C3A cells were seeded on the membranes with micropattern of 200μm in spacing and 80μm in depth, most of cells adhered and aggregated in the grooves on the membranes in a few minutes. During the course of cell culture, the cells could differentiate, proliferate, and migrate up to the ridges. The cell growth and metabolic rates on the micropatterned CCG membranes were much higher than those on the T-flask well. It indicated that the micropattern on the membrane surface could affect the cell distribution and communication, and resulted in difference of cell activities in adhesion, morphology, cell growth, and mobility.
