本研究探討以固相萃取法及石墨爐式原子吸收光譜儀測定豬肉中血質鐵之含量。其方法為將豬肉以80%酸性丙酮溶液(pH<1)均質萃取,取上澄液10 ml加水10 ml,通過碳18處理匣,血質可被吸附,再以80%丙酮溶液5 ml沖提,將沖提液以偏極化茲曼石墨爐式原子吸收光譜儀直接測定其吸光度,由標準曲線計算其含量。添加含血質鐵1.75、2.50及3.50μg/g之肌紅素於豬肉中,回收率為97.9、98.1及99.2%;添加含血質鐵3.00、7.00及10.00μg/g之血紅素於豬肉中,回收率為98.1、98.8及99.3%。
A new method of selective determination of heme iron in pork was developed. First heme iron in pork was extracted with 80% acetone acidified with 2% hydrochloric acid to below pH 1.0.After adjusting the concentration of acetone to 40%, the solution was passed through a Sep-Pak C18 cartridge. Heme was adsorbed on the cartridge, and was then eluted with 80% acetone. The eluentwas analyzed for heme iron with a graphite furnace atomic absorption spectrophotometer. Recoverys tudies were performed by adding 1.75, 2.50 and 3.75 μg HFe/g myoglobin and 3.00, 7.00 and 10.00μg HFe/g hemoglobin into the pork. Recoveries were in the range of 97.9~99.3%.9.3%.
