中文摘要

     目的:芳香胺化合物經N基乙醯轉移酵素之N基乙醯化後可誘發大腸癌,本研究是in vitro 探討正常大腸組織及大腸癌組織中乙醯轉移酵素的活性及 Vitamin E 對正常大腸組織及大腸癌癌組織中乙醯轉移酵素活性的影響。材料與方法:取 40 個正常大腸組織及大腸癌組織利用乙醯化輔脢作乙醯基而再利用高壓層析儀分析乙醯轉移酵素的活性及 VitaminE 對正常大腸組織及大腸癌癌組織中乙醯轉移酵素活性的影響, 以 aminofluorene 和p-aminobenzoic acid 作受質。結果:結果大腸癌組織乙醯轉移酵素活性較正常組織中乙醯轉移酵素活性高,Vitamin E 對正常大腸組織及大腸癌癌組織中乙醯轉移酵素的活性有降低的作用。 結論:Vitamin E 對大腸癌的治療及預防有可能具臨床的價值。本實驗報告是 invitro 探討的初步報告,當然須再進一步 in vivo 的研究確認。

英文摘要

     Purpose:Arylamine N-acetylation capacity by the N-acetyltransferase( NAT ) may be an important causative factor in the initiation of colorectalcancer. Vitamin E ( alpha-tocopherol ) has been known for many years as anantioxidant and act as an important component of anticancer regiments. Theactivity of NAT and the effect of vitamin E on NAT activity in healthy colon andcolon cancerous tissues ( invitro ) were investigated.Materials and Methods:Forty human healthy colon and colon cancerous tissue cytosol ( subcellularfraction ) were investigated for NAT activity. The effect of vitamin E on theactivity of NAT was also examined by high pressure liquid chromatography ( HPLC) using 2-aminofluorene and p-aminobenzoic acid as substrates in this study.Ressults: NAT activity are higher in colon cancerous tissue cytosol than inhealthy colon tissue cytosol vitamin E could decrease NAT activity in healthycolon and colon cancerous tissue cytosol. Conclusion:The results may suggest aclue to the use of vitamin E in treatment and prevention of human colon cancer.In future, in vivo study will be investigated.
