本研究以東方型雜交百合(Lilium spp. ‘Casa Blanca’)為材料,探討低功率發光二極體(Light emitting diodes;LED)組合光源對組織培養苗增殖與後續發根之影響。利用三種波長 LED 為組織培養的光源,設計六種LED 光源組合:紅光 (R)、藍光 (B)、紅加藍光 (RB)、紅加遠紅光 (RFr)、藍加遠紅光 (BFr) 和紅加藍加遠紅光 (RBFr),並以傳統螢光燈管 (FL)為對照組,每日照光16 小時、黑暗8 小時。以東方型雜交百合小鱗片為培植體,培養於 MS 添加 0.1 mg·L-1 NAA 與 1.0 mg·L-1 BA 之誘導鱗片增殖培養基中35 天後,照射 RFr、B 兩組之鱗片增殖數較高。以MS 基本配方培養時,照射 RBFr 與 RB 兩組之組培苗鮮重、乾重較大且總根數較多。將小鱗片微扦插於 MS 培養基中, 照射 RBFr 12 週後具有較佳之組織培養生長效果。遠紅光需同時組合紅光與藍光光源,方可促進鱗片葉形成與提高不定根數,進而達到東方型雜交百合瓶內生長效果。

英文摘要

In vitro multiplication and root formation stages of lily scale segments were studied under different light qualities including fluorescent (FL), blue LED (B), red LED (R), red plus blue LED (RB), red plus blue and far-red LED (RBFr), red plus far-red (RFr), and blue plus far-red (BFr) under 16 h light and 8 h dark photoperiod per day. RFr and B irradiation resulted in greater scale multiplication when inoculated with MS supplemented with 0.1 mg·L-1 NAA and 1.0 mg·L-1 BA for 35 days. The fresh weight, dry weight, and total roots of lily explants were higher with RBFr and RB treatments when inoculated with MS without growth regulator supplements. In root formation stage, best growth was found with RBFr treatment after 12 weeks.Results suggest that far-red in combination with red and blue light was required to enhance the scale-leaves and rooting.
