中文摘要

本研究探討石蓮花水與50%、70%、95%乙醇萃取物之抗氧化、及抗突變效應。利用Ames test評估石蓮
花萃取物之抗突變效果,結果顯示各萃取物濃度於1~10 mg/plate範圍內對TA 98與TA 100菌株無細胞毒性與致突變性,且對直接致突變劑NQNO具有抗突變效果 (可達28%),但對需經肝臟酵素代謝活化之間接致突變劑B(a)p並不具明顯抑制效果。抗氧化能力試驗方面,石蓮花各溶劑萃取物清除 DPPH自由基能力呈劑量效應,濃度於10 mg/mL下,清除能力皆可達80%以上,還原力及總抗氧化能力均以50%及70%乙醇萃取物較佳,但水萃取物對超氧陰離子則可展現較佳之清除活性,當濃度於5 mg/mL時,可達80%以上。除此之外,進一步分析其抗氧化活性成份,發現總酚化合物與類黃酮含量以95%乙醇萃取物最高,分別為33.0 mg/g與17.0 mg/g。綜合以上結果顯示,石蓮花係為安全且具抗氧化能力之天然植物,未來可開發成多項具保健功效之產品。

英文摘要

This study investigated that the antimutagenicity and antioxidative activities of Graptopelalum paraguayense that was extracted using water and various concentrations of ethanol (50%, 70%, and 95%).
As analyzed on Salmonella typhimurium TA98 and TA100 on Ames test, the results indicated that all extracts at the concentration of 1~10 mg/plate showed no cytotoxicity and mutagenicity and antimutagenicity up to
28% against direct mutagen NQNO without S9 enzyme, but there’s no effect on indirection mutagen benzo(a) pyrene (B(a)P) with S9 enzyme. Furthermore, all extracts of Graptopelalum paraguayense at concentration
of 10 mg/mL showed the DPPH scavenging activity up 80% and in dose-dependent manner. The 50% and 70% ethanolic Graptopelalum paraguayense extracts showed highest antioxidant activity on reducing power
method and trolox equivalent antioxidant capacity (TEAC). Another, the water extract at the concentration of 5 mg/mL showed high value of scavenging activity on superoxide anion up to 80%. Furthermore, the 95%
ethanolic Graptopelalum paraguayense extract were found to have highest total phenolic compounds and isoflavonoids which were 33.0 mg/mL and 17.0 mg/mL respectively. It was concluded that Graptopelalum paraguayense was a safe and had antioxidative activity that it could be developed as functional product.
