本研究以烏骨雞爪作為試驗原料,分別使用三種酵素:Alcalase(A)、Protease N(P) 和Flavorzyme(F),以兩階段方式進行水解作用,並將所得之三種水解物 (AF、PA及PF組) 經冷凍乾燥後分析其可溶性蛋白、胜肽及黑色素含量、紫外線吸收光譜、以及抑制酪胺酸酶能力,並將總評效果最佳之組別進行胺基酸組成分析,進而評估以兩階段酵素水解烏骨雞爪製備活性胜肽之可行性。結果顯示,烏骨雞爪經過不同酵素的兩階段水解後,各組水解物中之可溶性蛋白質含量皆顯著降低,而胜肽含量則顯著增加 (p < 0.05),顯示各酵素組合均具有良好之水解效果;而在紫外線吸收光譜方面,所有烏骨雞爪酵素水解物在紫外線的波長範圍內皆有良好之吸光能力;其中PA組於UVA範圍內的吸光能力高於其他組別;在黑色素含量方面,每克水解物中分別含有1.92-2.32 mg之黑色素;此外,烏骨雞爪酵素水解物皆具有抑制酪胺酸酶能力,其中以PA組有顯著最佳的抑制效果。烏骨雞爪酵素水解物具有能用來作為化粧品中抑制酪胺酸酶潛力的細胞光保護成份。

英文摘要

The aim of this study was to investigate the functional characteristics of two-stage enzymatic hydrolysates from silky fowl feet treated with Alcalase (A), Protease N (P), and Flavorzyme (F). Three combinations, AF,
PA and PF, were used. After hydrolyzing for 4 hours, the hydrolysates were lyophilized and analyzed for the soluble protein, the peptide content, UV absorption spectrum, the melanin content and the ability to inhibit tyrosinase activity. The results showed that after enzyme treatment, the soluble proteins and peptide contents of the hydrolysates were decreased and increased, respectively. This result indicates that all the enzyme combinations possess good hydrolyzing ability. All the hydrolysates had UV absorption and tyrosinase inhibitory activity (PA had the highest absorption and tyrosinase inhibitory activity). In the melanin content analysis,three hydrolysates contained about 1.92-2.32 mg/g melanin. Silky fowl feet hydrolysates have high potential
