中文摘要

氧化壓力是造成細胞成分傷害的主要因素。本文旨在探討愛玉子種子水萃物 (JF萃取物) 對氧化壓力所誘導之大鼠肝臟Clone 9細胞損傷的保護作用。以四氮唑鹽 [3-(4,5dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide, MTT] 和瓊脂膠體電泳比色法分析,結果顯示JF萃取物能降低由過氧化氫誘導之去核醣核酸和Clone9細胞之損傷,而此種降低效果與所使用之JF萃取物濃度有關。此外,經過0.5 mM過氧化氫和100 μg/mL愛玉子種子水萃物處理的Clone 9細胞,其細胞型態與控制組並無差異。利用流式細胞儀分析發現,預先以100 μg/mL愛玉子種子水萃物處理,可顯著降低過氧化氫誘導之Clone 9細胞凋亡 (Sub G1) 比例和caspase-3活性。顯示愛玉子種子水萃物具有開發成為預防氧化壓力造成之失調疾病藥劑的潛力。

英文摘要

Oxidative stress is a major cause of injury to cellular components. In this study, the effects of aqueous extract from the jelly fig seeds (JF extract) on the prevention of oxidative stress-induced damage in rat
hepatocyte Clone 9 cells were investigated. MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay and agarose gel electrophoresis analysis showed that the JF extract was able to attenuate the oxidative stress damage induced by H2O2 in a concentration dependent manner. In addition, no morphological difference was observed between the control and the Clone 9 cells treated with 0.5 mM H2O2/100 μg/mL JF
extract. The pretreatment of Clone 9 cells with JF extract has successfully prevented H2O2 damage as shown in the reduction in percentage of apoptotic cells (SubG1 peak) and the caspase-3 activity in flow cytometry.These evidences revealed that JF extract has the potential to be a prophylactic agent in the prevention of oxidative stress-mediated disorders.
