目前對於美白成分的篩選方法,主要是利用抑制酪胺酸脢活性來判定其美白功能,並利用檢測OD475nm的吸光度,但是利用此種方法可能面臨一些問題,由於一些中草藥在萃取時都會有一些植物色素的殘留,在進行酪胺酸活性抑制檢測時會影響吸光值的偵測。另外蛋白質體學可以綜觀細胞內全體蛋白質之變化,建立種方法將有助於研究美白成份對細胞之生化代謝影響。所以此研究主要是建立酪胺酸脢活性染色方法,避免植物色素干擾美白成份之篩選,並且建立黑色素細胞蛋白質體分析方法,以一較全面性研究方法,進行植物美白成份篩選並研究其對細胞之整體影響。
The most commonly used method for screening skin lightening agents is to detect tyrosinase inhibition by measuring the absorbance at OD 475 nm. This method could face difficulty, when the testing subjects such as Chinese herbal extracts contain colored compounds which would interfere with the measurement of the absorbance. Proteomic analysis of cell protein provides a overall view of protein changes in the cell and could help investigate the effect of skin lightening agent on the biochemical metabolism of cell. This study is to establish a method of tyrosinase activity stain for evaluating skin lightening agents without the interference of colored compounds and to establish a method of proteomic analysis of Melanocyte cell for evaluating skin lightening agents from plants and studying their overall effect on cell.
