本研究使用Inter Simple Sequence Repeat (ISSR)分子標誌分析長壽花品種間之遺傳歧異度。試驗以29 個長壽花品種及落地生根與鵝鑾鼻燈籠草為材料,所篩選12 條ISSR 引子,共可擴增出179 個條帶。以Jaccard’s coefficient 計算品種間遺傳相似度介於0.165-0.74 之間。以Unweighted Pair Group Method with Arithmetic Mean(UPGMA)進行群聚分析及主成份分析的結果相似。在遺傳相似度0.42 處可以將長壽花分為兩群;第一群是經種間雜交所獲得具有鵝鑾鼻燈籠草遺傳背景之2 品種,而第二群為種內雜交的27 個品種。而在遺傳相似度0.495 處又可以將第二群長壽花品種再劃分為兩小群。相同公司所育出的品種有歸屬同一群的趨勢。
In this study, Inter Simple Sequence Repeat (ISSR) fingerprinting was employed to investigate the genetic diversity of kalanchoe cultivars. Twenty-nine kalanchoe cultivars plus K. garambiensis and K.pinnatum as out-groups were involved in the study. The genetic diversity was analyzed by using 12 selected ISSR primer pairs and 179 dominant polymorphic markers identified from the kalanchoe cultivars. The genetic similarity calculated according to Jaccards’s coefficient was between 0.165 and 0.74. Results obtained from the Unweighted Pair Group Method with Arithmetic Mean (UPGMA) analysis and the principle component analysis was similar. Twenty-nine kalanchoe cultivars could be clustered into two
groups separating at the genetic similarity of 0.42. One group containing only two cultivars both shared common genetic background of K.
garambiensis seeming derived from inter-species crosses. The other group
of 27 cultivars derived from intra-species crosses. The second group of kalanchoe cultivars could be further clustered into two sub-groups separating at the genetic similarity of 0.495. Cultivars developed by the same company always fell into the same group and could be easily distinguished.
