中文摘要

本研究探討桑黃 (Phellinus linteus) 於5-L攪拌式生物反應器生成菌絲體生質量及生理活性物質(β-1,3-glucan、總酚)之培養條件。試驗結果顯示,P. linteus生成菌絲體的較適接種量、通氣量與攪拌速度分別為5%、1.5 vvm與125 rpm。P. linteus於上述較適條件培養20天,其最大菌絲 生成量、菌絲體生成速率與菌絲體產率分別為8.86 g dry mycelial biomass/ L、0.44 g dry mycelial biomass/ L/ day與0.44 g/g glucose,與培養於基礎發酵條件 (接種量為10%、通氣量1.0 vvm與攪拌速度100 rpm) 比較,培養於較適條件之最大菌絲體生成量為基礎發酵條件的1.93倍,菌絲體生成速率與菌絲體產率則為1.91倍。在20天發酵期間發酵液之總酚含量與總抗氧化能力隨P. linteus菌絲體生成量增加而增加,但是β-1,3-glucan濃度隨培養時間增長而減少。

英文摘要

The influences of cultivation conditions on mycelial biomass and bioactive compounds produced by a medicinal mushroom Phellinus linteus in a 5-L stirred-tank bioreactor were studied. The results showed that the optimal inoculum ratio, aeration rate and agitation rate for the production of mycelia biomass were 5%, 1.5 vvm and 125 rpm, respectively. Then, the mushroom was grown under the optimal cultivation conditions (inoculum ratio 5%, aeration rate 1.5 vvm and agitation rate 125 rpm) in a 5-L stirred-tank bioreactor after 20 days of incubation time, and the maximal production of mycelial biomass, mycelial biomass formation rate and mycelial biomass yield were 8.86 g/L, 0.44 g/L/day and 0.44 g/g glucose, respectively. These results were 1.93-, 1.91- and 1.91-fold of the maximal production of mycelial biomass, mycelial biomass formation rate and mycelial biomass yield of P. linteus under the basal cultivation conditions (inoculums ratio 10%, aeration rate 1.0 vvm and agitation rate 100 rpm). During 20 days of incubation time, the total phenolic content and values of Trolox equivalent antioxidative capacity for P. linteus culture broth were increased with production of mycelia biomass. However, the content of β-1, 3-glucan was decreased during incubation time.
