生物快速篩檢法 (DR CALUX®) 屬於一種應用報導基因的檢測技術,主要是利用活化多環芳香烴受體並啟動冷光報導基因的表現來檢測戴奧辛類化合物。本研究建立水產動物基質之DR CALUX®生物快速篩檢法並應用於多種魚類、雙貝類及甲殼類的戴奧辛類化合物檢測。水產動物經萃取後,包括石斑魚、七星鱸魚、鱔魚、文蛤、牡蠣及白蝦等六種水產動物的平均脂肪含量小於3.0%,其餘樣品的脂肪含量範圍為4.5%-28.4%。DR CALUX®生物快速篩檢法應用於魚肉樣品的偵測極限值為0.01-0.04 pg DR CALUX-TEQ/g w.w.,定量極限值為0.02-0.15 pg DR CALUX-TEQ/g w.w.;而雙貝類及白蝦的偵測極限值為0.01 pg DR CALUX-TEQ/g w.w.,定量極限值為0.02-0.03 pg DR CALUX-TEQ/g w.w.。水產動物樣品的戴奧辛類化合物經檢測結果顯示魚肉樣品的總毒性當量範圍為0.04-1.24 pg DR CALUX-TEQ/g w.w.,雙貝類為0.13-1.43 pg DR CALUX-TEQ/g w.w.,蝦類為0.05-0.14 pg DRCALUX-TEQ/g w.w.,所有樣的戴奧辛含量皆低於歐盟訂定的水產動物中 (不包含鰻魚) 戴奧辛類化合物之最大限值8.0 pg WHO-PCDD/F-PCB-TEQ/g w.w.。

英文摘要

The DR CALUX® (Dioxin Responsive-Chemical Activated LUciferase eXpression) bioassay is a type of reporter-gene assay that can detect dioxin-like compounds by activating the aryl hydrocarbon receptor (AhR),
causing expression of the reporter gene. This study built up the DR CALUX® bioassay to determine the toxicity equivalent (TEQ) of dioxin-like compounds (PCDDs/DFs/dl-PCBs) in aquatic organisms and the assay was
applied to determine the dioxin-like compounds in many species of fish, two species of bivalves, and one species of shrimp. The averages of the fat content of grouper, swam eel, hard clam, oyster, and white shrimp
were less than 3.0%, and those of other samples were between 4.5% and 28.4%. The LOD and LOQ of the DR CALUX® bioassay for fish was 0.01-0.04 pg DR CALUX-TEQ/g w.w. and 0.02-0.15 pg DR CALUX-TEQ/g w.w.,and for bivalves and shrimp they were 0.01 pg DR CALUX-TEQ/g w.w. and 0.02-0.03 pg DR CALUX-TEQ/g w.w., respectively. The total levels of dioxin-like compounds in fish samples ranged from 0.04 to 1.24 pg DR CALUX-TEQ/g w.w., in bivalve samples ranged from 0.13 to 1.43 pg DR CALUX-TEQ/g w.w., and in shrimp samples ranged from 0.05 to 0.14 pg DR CALUX-TEQ/g w.w.. These levels are all below the European Union’s approved maximum limit (8.0 pg WHO-PCDD/F-PCB-TEQ/g w.w., with the exception of eel) for seafood.
