使用acetone作為萃取溶劑,成功從CYY95-26甘藷中萃取β-carotene,使用acetone的萃取效果也優於ethanol及methanol,會影響萃取效果的因素包括萃取時間與acetone濃度兩個變數,根據反應曲面的實驗設計法(RSM),acetone濃度對萃取液的吸光值有顯著的影響,而迴歸model所建議的最適化萃取條件為20.6 min與使用95.8% acetone,若加入4 min超音波處理,可以提高β-carotene的萃取效果。CYY95-26甘諸acetone萃取物清除DPPH自由基的EC50值為0.39mg/ml,經與文獻中其他品種甘藷的EC50值比較,CYY95-26甘藷清除DPPH 自由基的效果較佳,其抗氧化能力也較好。
β-carotene was successfully extracted from CYY95-26 sweet potatoes using acetone as extraction solvent. Results from this study showed that using acetone achieved a better extraction performance than ethanol and methanol. The extraction time and concentration of acetone were two variables that affected the β-carotene extraction process. The degree of acetone concentration had a significant impact in determining the absorbance of extracted β-carotene solutions. Based on the regression model established by the response surface methodology (RSM), the optimal extraction conditions for CYY95-26 sweet potato β-carotene extraction were 20.6 min of extraction time and the use of a 95.8% concentration of acetone. The extraction performance was enhanced by introducing 4 min of sonication treatment. The EC50 value of the CYY95-26 sweet potato acetone extract was 0.39 mg/ml. Comparison with previous study results indicated that CYY95-26 has higher antioxidant capacity and higher DPPH radical scavenging activity than other sweet potato varieties.
