加馬射線是高能量生物傷害的游離輻射,食品科學應用於保持植物種子新鮮免於蟲害、黴菌。本研 究以不同劑量加馬射線照射綠豆,觀察不同劑量照射之綠豆其植株生長情形、脂質過氧化、類黃酮化合物 含量及自由基清除能力。利用蛋白質體學方式研究綠豆接受不同劑量之加馬射線照射時其蛋白質變化情 形,比對植物蛋白質序列資料庫後,發現輻射照射傷害可能誘導表現蛋白質,以此探討游離輻射傷害對植 物蛋白質之影響。結果顯示較高劑量加馬射線照射種子其植株發育不佳、脂質過氧化情形較低、類黃酮化 合物含量及自由基清除能力提高,由差異性蛋白質中發現5個與游離輻射傷害相關蛋白質,藉由調控植物 生長發育與代謝對抗游離輻射傷害。

英文摘要

Exposure to gamma ray irradiation is a frequent, clean, and superior method used to prevent bacterial fungi contamination of sterilized foods. In this study, the Vigna radiata was expressed to different dose gamma ray irradiation. The concentration changes of malondialdehyde, flavonoids and the DPPH radical scavenging activity after gamma ray irradiation on Vigna radiata were measured by ELISA method. To enhance our understanding of Vigna radiata proteome after gamma ray irradiation, the proteins were analyzed using proteomic approaches followed by peptide fragmentation patterning. For the experimental results, the malondialdehyde was down-regulated, flavonoids and the DPPH radical scavenging activity were up-regulated after gamma ray irradiation. Also, five proteins with differential expression were identified and related with and may represent proteins specific for ionized radiation damage. Those proteins may affect the plant growth, development and metabolism.
