中文摘要

阿維菌素類抗生素 (Avermectins) 屬於巨環內酯類抗寄生蟲藥,其優異的抗內外寄生蟲活性常用於畜產動物,由於該類藥物屬於非極性,在動物體內分佈性大且半衰期長,因而形成此等藥物在動物體內可食性組織的殘留問題,故需加強本類藥物的檢測。本研究以液相層析串聯質譜儀,使用電灑游離正離子法,可同時檢測檢體中阿巴汀 (abamectin)、多滅蟲 (doramectin)、eprinomectin、emamectin、愛滅蟲 (ivermectin) 及moxidectin,六種avermectin類抗生素。本試驗檢體以乙腈萃取,選用Sep-Pak C8之固相萃取匣進行樣品淨化,沖提液以去離子水稀釋定容至10 mL上機,採用xselect HSS T3層析管柱,移動相為0.1%甲酸水溶液與乙腈,以液相層析串聯質譜儀進行分析。肉品類添加抗生素後平均回收率介於53.1-91.8%,同日內平均變異係數介於0.2-11.6%,異日間之平均變異係數介於0.3-12.3%間,定量極限介於0.5-10 ng/g;內臟類檢體平均回收率介於60.6-89.6%,同日內平均變異係數介於0.2-8.8%,異日間之平均變異係數介於0.7-13.6%間,定量極限介於1-25 ng/g;基質匹配檢量線的迴歸係數 (R) 皆達0.995以上。

英文摘要

Avermectins belong to a group of compounds called the macrocyclic lactones. Their excellent resistance to endoparasitic and ectoparasitic infections enables them to be commonly used in food-producing animals. Avermectins are lipophilic and have long half-life, which results in the drug residue problem in edible tissues of animals. Liquid chromatography tandem mass spectrometry using an electrospray ionization positive ion method can simultaneously detect avermectins, such as abamectin, doramectin, eprinomectin, emamectin, ivermectin and moxidectin. The analyte was extracted with acetonitrile, and cleaned up using the Sep-Pak C8 solid phase cartridges. The eluent was diluted with deionized water to 10 mL. The analysis was performed on a XSelect HSS T3 column with mobile phase of 0.1% formic acid and acetonitrile. The average recoveries of avermectins spiked in meat ranged between 53.1-91.8%; intra-day coefficients of variation ranged between 0.2- 11.6%; interday coefficients of variation ranged between 0.3-12.3%; the LOQ were in the range between 0.5-10 ng/g. The average recoveries of avermectins spiked in viscera ranged from 60.6-89.6%; intra-day coefficients of variation ranged from 0.2-8.8%; coefficients of variation of the inter-day ranged from 0.7-13.6%, LOQ ranged from 1-25 ng/g. The regression coefficients (R) of all the matrix-matched calibration curves are above 0.995.
