當歸、川芎、熟地和芍藥 (俗稱四物) 及丹參為傳統的中藥素材,在亞洲國家常用於治療婦科疾病。超音 波輔助萃取技術相較於傳統萃取法成本相對較低,操作簡單、效率高,應用於天然植物有效成分之萃取已相 當普遍,但對於丹參四物飲品萃取的研究仍相當有限,因此本試驗欲藉由超音波輔助萃取,以分光法測定總 酚和皂素並以高效能液相層析質譜法 (High-performance Liquid Chromatography-Mass Spectrophotometry, HPLC-MS) 分析沒食子酸、阿魏酸、芍藥苷、5-羥甲基糠醛、丹參酚酸B及丹參酮IIA等含量,以探討四物及丹參最適萃 取參數。此外,利用多光源背相散射光 (turbiscan) 量測有無超音波處理的四物飲穩定性。結果顯示,四物最適 處理條件為乙醇濃度20%、固液比1 : 10、萃取溫度50℃ 及處理四小時;丹參最適萃取條件為20%乙醇、固液比 1 : 10、萃取溫度50℃ 及萃取一小時;經超音波萃取丹參四物飲的沉澱量變少,穩定性明顯提高。綜合以上結 果,超音波高頻震盪能微細化萃出物,有助於提升其穩定懸浮性,由於國內四物飲產品並無統一規格,而本 試驗以機能性成分作為標的,有助於建立相關保健產品的品質規格。

英文摘要

Angelica sinensis, Ligusticum chuanxiong, Paeonia lactiflora, Rehmannia glutinosa (Si Wu) and Salvia miltiorrhiza (Danshen) are traditional Chinese medicine used to treat women’s diseases in Asia. Ultrasound assisted extraction is widely used for nature active compounds because its relatively low cost, simple operation and high efficiency compare to traditional extraction methods. However, studies of Danshen-Si Wu drink are still limited. Therefore, the aim of this study was to investigate the optimal ultrasound-extracted parameters of Si Wu and Danshen. We not only analyzed total phenolic compounds and saponins by spectroscopy but also gallic acid, ferulic acid, paeoniflorin, 5-hydroxymethylfurfural (5-HMF), salvianolic acid B and tanshinone IIA by high-performance liquid chromatography-mass spectrophotometer (HPLC-MS). In addition, we also measured the stability of Si Wu treated with or without ultrasound by turbiscan. The results showed that the optimal extracted parameters of Si Wu were as follows: 20% aqueous ethanol; ratio of material to solvent: 1 : 10 (w/v, g/mL); extraction temperature: 50oC; extraction time: 4 hr. The optimal extracted parameters of Danshen were as follows: 20% aqueous ethanol, ratio of material to solvent: 1 : 10 (w/v, g/mL); extraction temperature: 50oC, extraction time: 1 hr. Under ultrasound extraction, its precipitation reduced and stability improved. In conclusion, ultrasound extraction could refine the Danshen-Si Wu drink extract and elevate its suspension stability. Because there is no unified Si Wu format in Taiwan, this study may by helpful for establishment of the quality format of related functional products on the basis of their active compounds.
