中文摘要

氧化壓力所造成的精子損傷是男性不孕症的主因之一,因此抗氧化劑與男性不孕之關聯已有諸多文獻探 討。研究指出薑黃素具有抗氧化及抗發炎等生理活性,然臨床數據顯示其生物利用率極低,且為相當不安定 的化合物。本研究以二氧化矽與幾丁聚醣包埋薑黃素製成奈米化薑黃微粒 (Nanocurcumin, NACU),探討其對順 鉑 (Cisplatin, CP) 誘導倉鼠生殖能力損傷之修復效果。結果發現適當濃度之NACU能有效增加精子濃度、活動 力並減少形態異常精子之比例,且可顯著降低因順鉑所誘導的倉鼠精子內ROS (Reactive oxygen species) 生成量 及血漿、睪丸和精子中脂質過氧化終產物MDA (Malondialdehyde) 的含量,證實了奈米化薑黃素可減少氧化壓 力對精子所造成的損傷,可作為日後預防男性不孕症的潛在保健品開發。

英文摘要

Sperm damage caused by oxidative stress is one of the main reasons for male infertility. Researches had shown that curcumin has many activities including anti-oxidation and anti-inflammation; however, the clinical data indicate that it is a very low bio-available and unstable compound. In this study, the curcumin nanoparticles (Nanocurcumin, NACU) fabricated by chitosan and silica were investigated for the effect of repair ability in a hamster reproductive capacity which damaged by cisplatin (CP). It was found that an appropriate dosage of NACU can effectively increase the sperm concentration, motility and reduce the proportion of abnormal sperms. NACU can also significantly reduce the excess ROS caused by CP in sperm and MDA content in plasma, testicular and sperm. We have clarified the ability of the nanocurcumin to repair sperm damage by reducing oxidative stress and NACU can be used as a potential health product to prevent male infertility.
