Camellia oleifera Abel.,俗名為油茶,其種籽含油量高,可壓製成食用油脂,為台灣民間所謂之油茶油。 其所餘之油茶粕含有豐富的皂素。雖然目前已有許多研究關於油茶籽皂素之活性與毒性的研究,但卻鮮有 研究關於油茶粕中類黃酮結構之相關研究。因此,本研究證實可利用FIA/ESI/Ion-Trap/MS此分析技術,建立 一套快速、省力、經濟及有效的類黃酮分析鑑定的方法。本試驗為油茶粕甲醇萃取物中類黃酮之分離與鑑 定,並探討油茶粕甲醇萃取物之抗氧化成份與活性。本試驗發現油茶粕甲醇萃取物中含有類黃酮,經質譜鑑 定,發現其中兩個主要類黃酮之分子量為756及726 Da.,其結構為kaempferol-3-O-hexose-deoxyhexose-hexose及 kaempferol-3-Openatose- deoxyhexose-hexose,推測可能為camelliaside A及B,但本研究所得結構之含糖種類及其 鍵結之位置,尚未明朗,故須進一步之探討之後才能確定。另外,以HPLC-DPPH.系統分析油茶粕甲醇萃取 物之抗氧化物質及其活性。結果發現,kaempferol之配醣體並無抗氧化能力,但將kaempferol之配醣體水解釋 出kaempferol後,則具有抗氧化能力。

英文摘要

Camellia oleifera Abel. is a kind of plant beings to tea family and is usually termed as Kuu-Char in Taiwan. Seeds of oil tea can be used to produce edible oils. The oil free residues are saponin-rich materials. Quite a few research groups have reported the bioactivity potency of oil tea saponins. However, to our knowledge, the structures of these flavonoids are still unclear and remained to be elucidated. Therefore, the purpose of the present study is to isolate and identify the flavonoids in oil tea seeds. Therefore, this study confirmed the use of FIA/ESI/Ion-Trap/MS analysis technology to establish a rapid, labor-saving, economical and effective method for the identification of flavonoids. In the preliminary studies, a group of components not belonging to flavonoids were found in the methanol extracts of the oil free residue of the seeds. The mentioned components were found to be flavonoids. Two of which with molecular weight 756 and 726 Da. were identified by ESI-MS as kaempferol-3-O-hexose-deoxy-hexose-hexose and kaempferol-3-O-penatose-deosyhexosehexose, respectively. Furthermore, the antioxidative components and activity of the methanol extracts were separated and evaluated by HPLC-DPPH device. The results showed that kaempferol glycosides did not have antioxidant capacity, but the kaempferol glycosides released kaempferol hydrolysis, it has antioxidant capacity.
