The aim of this study is to isolate, characterize, and identify lactic acid bacteria (LAB) from intestine of Pacific saury (Cololabis saira). A total of 33 Pacific saury samples were collected and 89 LAB were isolated. These isolates were divided into 11 groups (A to K) by using restriction fragment length polymorphism analysis and 16S ribosomal DNA sequencing. The most common bacterial genus found in Pacific saury was Pediococcus. On the basis of phylogenetic analysis of 16S rRNA, rpoA and pheS gene sequences, one Lactococcus strain were identified as potential novel species or subspecies. In addition, a total 6 isolates were found to have nisin- or pediocin-like bacteriocin producing abilities. Bacteriocin from strain Pediococcus acidilactici BTY-24-10 was further purified by using three purification steps. The molecular size was determined as 4613.021 Da which was close to that of pediocin AcM. These results showed that various LABs were found in the intestine of Pacific saury. This is the first report describing the distribution and varieties of LAB in the intestine of Pacific saury samples collected in Taiwan.
