香茹(Glossogyne tenuifolia, GT) 是傳統的中草藥具有抗發炎的活性。為了了解香茹對於免疫調節的能力,我們測試香茹對T 細胞誘發的適應性免疫力的影響。在本研究,我們發現香茹正己烷萃取液不會造成人類周邊血單核細胞(human peripheral blood mononuclear cells; PBMCs) 死亡,而且展現抑制人類周邊血單核細胞增殖及產生γ 干擾素(interferon γ;IFNγ) 的能力。進一步分析,我們發現香茹正己烷萃取液會抑制植物血凝素(Phytohaemagglutinin, PHA) 刺激人類周邊血單核細胞活化裂質素蛋白激酶(Mitogen-activated protein kinases, MAPKs),ERK2、p38 及JNK 的磷酸化。我們的研究顯示香茹正己烷萃取液藉著抑制MAPKs 的活性,調節T 細胞之活化。

英文摘要

Glossogyne tenuifolia (GT) is a traditional Chinese herbal medicine previously shown to exhibit anti-inflammatory activity. To explore the immune-regulating power of GT, we tested the effect of GT on T lymphocyte-mediated adaptive immunity. In this study, we obtained the n-hexane fraction of GT possessing suppressive potential in PHA-stimulating human peripheral blood mononuclear cells (PBMCs) proliferation and secretion of interferon γ (IFNγ). We demonstrate herein that the n-hexane fraction of GT inhibited T cell activation, which was not due to its cytotoxic activity. In addition, the n-hexane fraction of GT significantly reduced mitogen-activated protein kinases, extracellular signal-regulated kinases (ERK), p38 and c-Jun N-terminal kinases (JNK), phosphorylation in a T cell activation-dependent manner. Therefore, decreased MAPKs activation mediated the downrreguated effect of GT in T cell activation.
