順鉑(Cisplatin)為化療常使用的藥物之一,對於多種癌症有顯著療效,但順鉑也具有其副作用,包含噁心嘔吐、神經毒性及腎毒性等。本研究先以高劑量順鉑誘導急性腎損傷模式,於5株益生菌中篩選出最具有保護效果之益生菌Lactobacillus paracasei GKS6,結果顯示,給予500 mg/kg GKS6益生菌的尿素氮和肌酸酐可以降的最低。再以低、中、高劑量GKS6,評估對於慢性腎損傷之保護效果,結果顯示,給予125 mg/kg GKS6即可顯著降低尿素氮和肌酸酐含量(p<0.01),且具有劑量效應。於腎功能相關之發炎因子檢測中,給予125 mg/kg GKS6即可以顯著降低白蛋白、TNF-α、IL-1β、IL-6和TGF-β含量。綜合以上結果,證實GKS6對於腎臟保護具有良好的效果。
Cisplatin is commonly used in chemotherapy. It has significant effects on various cancers. However, cisplatin also has its side effects, including nausea, vomiting, neurotoxicity and nephrotoxicity. In this study, high-dose cisplatin was used to induce acute kidney injury, and the most protective probiotics were screened out of 5 probiotics. The results showed that the urea nitrogen and creatinine of 500 mg/kg Lactobacillus paracasei GKS6 probiotics can be reduced to the minimum. Then low, medium and high doses of GKS6 were used to evaluate the protective effect of chronic renal injury. The results showed that the administration of 125 mg/kg GKS6 can significantly reduce the content of urea nitrogen and creatinine ( p < 0.01), and has a dose effect. In the detection of inflammatory factors related to renal function, administration of 125 mg/kg GKS6 can significantly decrease the contents of albumin, TNF-α, IL-1β, IL-6 and TGF-β. Based on the above results, it is confirmed that GKS6 has a good effect on kidney protection.
