Wistar雄性幼鼠日齡14天時複製單側隱睪模型。在日齡28、50和100天時殺死,取兩側睪丸。日齡配對的未手術維和模擬組作對照組。觀察睪丸重量、容積、精細管平均直徑(Mean Seminiferous Tubule Diameter,MSTD)、睪丸活檢評分(Testicular biopsy score, TBS)和流式細胞計數(FLOW Cytometry, FCM)生殖細胞DNA分析。日齡28天和50天時對側己降睪丸(Contralateral Descended Testis, CDT)的重量、容積、MSTD和TBS與兩個對照組左側睪丸無顯著差異(P>0.05),日齡100天時CDT的重量和容積與兩個對照組左側睪丸無顯著差異;CDT的MSTD 253.5 ± 36.1 µm,TBS 3.65 ± 0.52分;未手術組和模擬組左側睪丸的MSTD 303.3 ± 17.7 µm和 303.0 ± 16.6 µm,TBS 9.58 ± 0.24分和9.6 ± 0.14分,CDT的MSTD和TBS與兩個對照組有顯著差異(P<0.01)。FCM DNA 分析,日齡28天時CDT的兩倍體細胞群(diploid,2C) 30.08 ± 9.97%;未手術組左側睪丸2C 75.57 ± 1.76%,兩組有顯著差異(P<0.01)。日齡50天和100天時CDT的單倍體細胞群(haploid,1C) 55.3 ± 7.69% 和52.4 ± 3.28%,未手術組左側睪丸的1C 64.39 ± 10.42% 和 60.42 ± 3.45%,兩組有顯著差異(P<0.02和P<0.01)。因此,Wistar鼠單側隱睪CDT有繼發性病變。

英文摘要

Unilateral cryptorchidism model was induced in Wistar rat at the age of 14 days. A shamoperative and A non-operative group were in closed control groups. Animals were sacrificed at 28, 50 and 100 days old, respectively. The weight and volume of testes, mean seminiferous tubule diameter (MSTD), testicular biopsy score (TBS) and DNA of germ cells were recorded. At the ages of 28 and 50 days, there was no difference in weight and wolulme of testes, MSTD and TBS between the contralateral descended testis (CDT) and the left testis of two control groups (P>0.05). At the age of 100 days, there was no difference in weight and volume between CDT and the left testis of two control groups, But the MSTD of CDT was 253.5 ± 36.1 µm, TBS was 3.63 ± 0.52 points; In the left testis of the non-operative and sham-operative groups, MSTD was 303.3 ± 17.7 µm V303.0 ± 16.6 µm; TBS was 9.58 ± 0.24 point V9.60 ± 0.14 points. There was a significant difference in MSTD and TBS between CDT and the left testis the two control groups (P<0.01). Using flow cytometry (FCM) to measure changes in DNA of germ cells, at the age of 28 days, diploid (2C) in CDT was 30.08 ± 9.97%; 2C in the left testis of the nonoperative group was 75.51 ± 1.76%, there was a significant difference (P<0.01). At the age of 50 and 100 days, haploid (1C) in the left testis of the non-operative group was 64.39 ± 10.42% V60.4 ± 3.45%respectively, In conclusion, these changes in CDT probable belong to the secondary degenerative lesion of cryptorchidism.
