本研究發現虱目魚皮明膠121oC熱水解物(retorted skin gelatin hydrolysates; RSGH)具有抗氧化活性,包括清除DPPH自由基能力、還原能力以及對DNA受過氧化氫之氧化傷害的保護效應。以不同熱水解時間(0.5~3 h)生成的抗氧化虱目魚RSGH (RSGH-0.5h, RSGH-1h, RSGH-2h, RSGH-3h),其中RSGH-3h有較佳抗氧化表現。以HPLC分析得知RSGH-0.5h, RSGH-1h, RSGH-2h, RSGH-3h主要分子量分布個別為6511 Da以上(62.48%)、686~12500 Da (87.49%)、189~6511 Da (81.39%) 與189~6511 Da (84.44%),較長時間熱水解促使RSGH生產更多水解胜肽和寡胜。本研究進一步將虱目魚RSGH-3h以超過濾(ultrafiltration, UF)依不同分子量篩選(1、5、10 kDa)劃分區分物為:1k-RSGH-3h(<1 kDa)、1-5k-RSGH-3h(1~5 kDa)、5-10k-RSGH-3h(5~10 kDa)與10k-RSGH-3h(>10 kDa)。結果顯示經UF之RSGH-3h提昇了其抗氧化性,其中1k-RSGH-3h有最好抗氧化表現,推測其因經高溫水解與超過濾處理,因而提高抗氧化胜肽濃度,顯示RSGH-3h中有許多分子量小於1 kDa的抗氧化寡胜肽存在。虱目魚水解胜肽產物1k-RSGH-3h,具有進一步研究潛力。

英文摘要

The present study demonstrates that milkfish retorted skin gelatin hydrolysates(RSGH) possess antioxidant activities including DPPH scavenging, reducing power, lipid peroxidation inhibition, and protective effect of DNA oxidative damage. The antioxidant activities of RSGH derived from different levels of thermal hydrolysis (from 0.5 to 3 h) showed skin gelatin retorted for 3 h (RSGH-3h) had the highest antioxidant activity. Besides, the main molecular weight ranges of the RSGH-0.5h RSGH-1h, RSGH-2h, and RSGH-3h were >6511 (62.48%), 686~12500 (87.49), 189~6511 (81.39%), and 189~6511 Da (84.44%), respectively. After sequential ultrafiltration with various MWCO ranges, 1k-RSGH-3h ultrafiltrated from RSGH-3h was the most strong antioxidant activity among 1k-RSGH-3h (MWCO < 1 kDa), 5-1k-RSGH-3h (MWCO: 1~5 kDa), 10-5k-RSGH-3h (MWCO: 5~10 kDa), and 10k-RSGH-3h (MWCO > 10 kDa). It indicated that oligopeptides in RSGHs play an important role in their antioxidant activities. Moreover, milkfish 1k-RSGH-3h shows great potential as a natural antioxidant.
